This proposal extends our synthetic program with oxazole-based marine metabolites to the structurally complex natural products phorboxazole A produced by the sponge Phorbos sp, and diazonamide A isolated from the sea squirt Diazona chinensis. These molecules are profoundly potent against a wide range of human tumour cell lines. Phorboxazole A has a structure based on a macrolactone core, four oxane and two oxazole rings and accommodates 15 asymmetric centres, 5E- and one Z-olefinic bonds. Our program will develop synthetic and stereo-selective methods to introduce these various functionalities, and incorporate sufficient flexibility to facilitiate their assemblage in the most flexible and economic manner. The structure of diazonamide A is based on a complex macrocyclic hetero-aromatic core which is further linked to a cyclopeptide portion. Our synthetic approach will feature the application of sequential sp2 - sp2 coupling protocols, including asymmetic variants of Heck and Ullman reactions, to elaborate the phenyl-indole, indole-oxazole, oxazole- oxazole, and quaternary carbon units in this novel and unusual structure.
