The proposed research will develop a catalytic intermolecular hydroacylation of enol ethers. The products of such a system are beta hydroxy ketones and are synthetically equivalent to the products of an aldol addition reaction. The hydroacylation approach is beneficial as it would be conducted under neutral conditions and should be mediated by low catalyst loadings - both significant advantages over the traditional aldol or Mukaiyama aldol reactions. The key to the success of the system is the use of aldehydes that bear chelating heteroatom groups. These chelating groups will allow the formation of stable acyl-metal intermediates.Initially, catalyst/substrate combinations will be sort that allow the simple (i.e. no diastero- or enantioselectivity considered) reaction to proceed. The system will then be extended to investigate the diastereoselectivity of the process by using both chiral aldehydes and chiral enol ethers. Ultimately an enantioselective variant of the process will be investigated. Applications of the methodology to the synthesis of polyacetate and polypropionate natural products will be explored.
