The project will involve development and survey applications of a high performance ion trap mass spectrometer: the instrument will provide a new tool for the screening of weak molecular interactions, such as metal-ligand and non-covalently bound biological complexes. The instrument will incorporate a liquid microjet interface, using a tunable and mid-infrared laser for desolvation and ionisation directly in the ion trap. The mass range of ions stored in the trap will be extended to higher values than those available in currently available commercial instruments by operating the ion trap RF field at lower frequency, use of resonant ion ejection, employing (pulse injected) helium buffer gas, and by the elimination of multipole transfer optics. High mass resolution, with good scan-to-scan reproducibility, will be achieved by employing the phase-locked mass-selective instability scan mode. The liquid microjet interface provides a powerful method for the detection of biological macromolecules as well as molecular ions of supra-, biomolecular and metal-ligand complexes in low charge states with a controllable degree of solvation. Our initial applications using this instrument will focus on studies of molecular recognition in multiplex oligonucleotide, protein, and metal-ligand complexes.
