Many of the procedures used to treat patients with occlusive arterial disease fail as a result of fibro-muscular thickening of the arterial wall at or near the site of endovascular or surgical intervention.In an effort to determine the extent to which haemodynamic factors contribute to the progression of this disease, a dynamic co-culture model of the blood-vessel wall is proposed. The model comprises a microporous, elastomeric conduit that is seeded with both endothelial and smooth muscle cells. Perfusion apparatus developed during the course of the project will be used to subject the above constructs to well-defined haemodynamic stresses representative of the vascular system in health and disease. The response of human smooth muscle cells to steady and pulsatile pressure and flow regimes will be assessed in terms of their proliferation, migration and capacity to synthesise extracellular matrix. Established immunohistochemistry and ELISA techniques will be employed to monitor the degree of interaction between these two cell lines and to study how endothelial cells communicate to the underlying smooth muscle cells the nature of the haemodynamic environment.
