The marine polycyclic ether toxins are important targets because of their structural complexity and putative biological activity. They represent a formidable synthetic challenge, especially with regard to the issues of ring construction and stereocontrol. An efficient total synthesis of any of these natural products will require the invention of new strategies and reactions for the construction of fused polycyclic systems. The proposed work will involve the synthesis of complex polycyclic systems in a novel, elegant and convergent manner using ring-closing metathesis as the pivotal reaction to effect ring construction. Ring-closing metathesis will be used to construct single-ring building blocks in enantiomerically pure form, and to prepare large polycyclic fragments by the union of these single-ring systems. This novel strategy will be based on our successful preliminary studies. The use of double ring-closing metathesis reactions for the two-directional synthesis of fragments of the polycyclic ether toxins will also be explored. This highly efficient strategy will be used to prepare tricyclic portions of some of the natural products in order to demonstrate the synthetic potential of this approach to polycyclic ether construction.
