The objectives of this proposal are the applications of ring closing alkene metathesis reactions in the total synthesis of complex b-lactam antibiotics. A series of polyfunctional dienes or enynes will be elaborated from simple precursors using sigmatropic rearrangements, cyclopropenylation or acyliminium ion chemistry. Subsequent reaction with metal carbenes such as Cl2(Cy3P)2Ru=CHPh should provide access to the corresponding bicyclic b-lactam antibiotics. The method should be applicable for diverse cephems, carbacephems, oxacephems, and the corresponding1-azabicyclo[5.2.0]nonan-9-one systems including novel compounds for bioassay against common bacterial pathogens including resistant organisms.
