The polarisation of 129-Xe nuclei can be increased by several orders of magnitude by means of optical pumping with laser light. The dramatically enhanced sensitivity in MR detection makes it possible to image the gas distribution in lungs and tissues. This project will develop techniques to study perfusion in tumours and healthy tissues by imaging hyperpolarised xenon dissolved in injectable solutions. Apparatus for the production of hyperpolarised xenon is already operating at the Institute of Cancer Research. The current light source allows production of xenon with polarisation of the order of 2-5%, sufficient for preliminary in vitro studies. The beam inhomogeneity and the broad linewidth of the laser source set severe limitations to the degree of polarisation attainable. We plan to increase the nuclear polarisation of xenon to over 50% by using a new generation of high power fibre coupled lasers which provide a more homogeneous beam and a narrower linewidth. This would allow us to extend our studies to volunteers and patients.
