This project aims to develop an efficient catalytic samarium (II) reagent system and to utilise such a system in the first catalytic samarium (II) mediated ketyl-olefin ring closure reactions. Ultimately, this novel catalytic chemistry, when applied to the wide range of samarium (II) mediated transformations, will make this chemistry applicable in new areas, such as the pharmaceutical industry (particularly process development) where its use might previously have been precluded due to the cost of stoichiometric quantities of the reagent. The utility of this novel chemistry will be illistrated in a concise route to (-)- stemoamide, a neurotoxin and powerful insecticide, which has been synthesised only twice before by rather laborious routes. In addition, the new methodology will be applied in an approach to the core of the structurally unique Pestalotiopsins, a new family of therapeutically promising natural products.
