We have recently investigated methods for the generation and use of vinylidene carbenes from ketiones. In appropriate circumstances the carbene insert into an adjacent C-H single bond to give a cyclic product. When this is not favourable a rearrangement take place to give an alkyne. In the first section of the project the insertion reaction is exploited to provide a rapid access to the zaragozic acid/squalestatin caore structure.Although the emphasis will be on the generation of chiral, non-racemic products using homochiral starting materials , the opportunity will be taken to study asymmetric catalysts in certain applications as a complementary method for asymmetric induction. There are no plans in this project to develop new ligands. We shall extend and improve this process to complete the synthesis. The second section of work depends upon the high energy of the carbene to generate a hindered system which acts as the precursor for a, a-disubstituted amino acids, a class of target which are otherwise difficult to prepare. In the final section of the proposal use is made of the rearrangement to form alkynes and cyclic enediynes via a ring expansion process.
