This application concerns the development of enzyme mimics by searching for a receptor for a transition state mimic, from a library of purely synthetic receptors, generated by a semi-rational combinatorial approach.Key features of this proposal are 1. The basic catalyst design, based upon our success in developing synthetic receptors able to bind peptides selectivity in water;2. The number of compound combinations that we will be able to screen - up to 100, 000, 000, 000;3. ability to screen for binding of the transition state and for binding of product and starting material;4. The use of high-loading beads to allow screening for catalytic activity on a single bead.Although this proposal is concerned with catalysis particularly for amide and ester hydrolysis, the strategies employed here should have general applicability to other systems. The development of synthetic molecules capable of mimicking enzyme behaviour will have considerable use in furthering our understanding of catalysis and binding in natural systems, but in the longer term will provide new catalysts for practical applications.
