This programme will develop and evaluate the construction of polymer-based u-TAS devices that are fully integrated. Liquid sample handling devices will be constructed to enable either reagent addition by electroosmosis or separation by isotachopheresis. The reacted or separated analyte species will be monitored by optical waveguide detection, fabricated on the same planar substrate. An injection moulding process will generate indented substrates which will subsequently be coated selectively to both control electroosmosis in the flow/separation channels and to fabricate the waveguides. An alternative procedure of using waveguide inserts will also be investigated. Waveguides will be based upon two complementary principles: the resonant mirror (RM) and the anti-resonant reflecting optical waveguide (ARROW). One significant advantage of injection moulding will be the ability to generate statistically significant samples of the devices, such that a realistic evaluation of device reproducibility will be possible. Although this programme aims to generate proof-of-concept devices, exploitation could lead to reusable hand-held measurement units into which the disposable polymer u-TAS components are located. These would in incorporate all the required chemistries for a wide range of applications involving both flow injection and separation techniques.
