We intend to apply previously developed IMDAF methodology to the total synthesis of phorbol. This will involve optimising and extending the synthetic sequence to the cyclopentneoid from Diels-Alder precursor and subjecting this to high pressure mediated cycloaddition followed by site selective epimerisation. Subsequent elaboration to the natural product (in the form of its low toxicity pentaol) requires conversion of the B ring carbonyl to a hydroxymethylcyclopropanation motif, stereocontrolled introduction of the C-12 hydroxyl group and dimethylcyclopropanation. This approach should also be valid for a range of phorbol derivatives and related daphnane structures.
