The aim of the proposed research is to investigate the practical generation of internal images of intact biological samples using two-photon excited fluorescence. Femtosecond, near infrared laser pulses will be used to excite blue-green fluorescence in tissue using two-photon absorption. The long excitation wavelength means that the internal structure of the samples can be optically sliced with minimal risk to the sample due to the low average power. To advance the technique from the laboratory bench to a system for applications research instruments will have to be developed. A compact, reliable, cost effective and simple to operate all-solid-state femtosecond laser source will be investigated and developed capable of imaging complete specimens.Using these instruments two-photon excited fluorescence images will be generated , non-destructively, of complete macroscopic biological samples. The initial investigation of the technique will be in dental and ophthalmic tissue and plant specimens. These have naturally present fluorophores that can be excited by the proposed laser system. The fundamental limits to the technique imposed by the scattering in the tissue of both the excitation and emitted light will be examined.
