An increasingly important strategy for active compound discovery in the fine chemicals industry involves the generation of large, diverse libraries of compounds for screening, by the matrix combination of reactive subunits. A common strategy for such a combinatorial library synthesis is to attach one subunit to a polymer bead, which is then sequentially exposed to different reactive subunits; the compounds generated are then cleaved from the support and screened. To advance this technology further, there is an urgent need for the development of : novel modes of attachment of subunits to the polymer; new reactions (hence product types) compatible with the polymeric support; and methods for cleavage from the support capable of introducing new functionality (and therefore diversity) at this step. The work to be undertaken in this project applies chemistry based on silyl diazocarbonyl compounds, developed in the investigators laboratories, to these problems. New silyl diazocarbonyl-bearing polymer supports will be prepared, and their diverse chemistry exploited to develop novel solid phase synthesise of a range of potential pharmacophores. The choice of silicon as a linking element allows the introduction of new functional groups on cleavage. The technology will be evaluated in collaboration with a major pharmaceuticals company.
