Summary on Grant Application Form
The primary aim is to orthogonal acceleration time-of-flight mass spectrometer for ultra-sensitive high speed liquid chromatography, in particular, capillary electrochromatography. Several pieces of enabling technology will be designed and developed by ion optical modelling and experimental prototypes, namely: 1) a system of fast ion-gates (comb filters) to increase the TOF duty cycle and hence the sensitivity, 2) the ion optical interface from the external ion source to the internal orthogonal source which will enhance sensitivity and mass resolution. The oa-tof analyser will be designed so that a cryogenic calorimetric detector can be fitted to greatly enhance the sensitivity of high m/z ion detection.Interfaces for electrospray, atmospheric pressure chemical ionization and nanospray will be developed for coupling to HPLC and CEC. Investigations on high speed chromatographic separations (peaks < 1 sec. wide) will be undertaken as well as nanospray studies of proteins in non-denaturing buffers requiring high m/z performance, for example, amyloid species.
