The study of co-operative interactions is a topic of great current interest because it is these interactions which determine the stability of DNA duplexes, of the folded states of proteins, of enzyme/substrate complexes, and of ligand/receptor interactions. The essentially universal principles which operate in co-operative binding can be illustrated by reference to the dimerisation of glycopeptide antibiotics, crucial in the treatment of resistant bacterial infections, are promoted in their activity via dimerisation. This dimersation is typically co-operative with the binding of bacterial cell wall analogues. A method is proposed to estimate what proportions of the free energy of the co-operativity arise at the dimer interface and at the ligand binding interface.
