The mechanism and structure of inositol monophosphatase, the putative target for lithium therapy, have been determined recently in our laboratory. A new range of mechanism-based inhibitors for inositol monophosphates will be synthesised by elaboration of 06-(2' -hydroxyethyl)-cyclohexane-1,2,4,6-tetraol phosphates and cyclic phosphates using methodologies starting from (-)-quinic acid. The compounds will be tested and the results will be used to test our understanding and to design and prepare highly specific compounds of potential medicinal value.
