A wide range of materials are used in the manufacture of prostheses. This combined with the diversity of surface treatments available, from continuous to porous coatings, surface modification of structure and topography means a diversity of cellular response occurs. The cellular responses are dependent on a range of parameters including, topography, surface structure and orientation, stoichiometric changes, surface changes and surface chemistry. These parameters will be targeted systematically by determining precise cellular and molecular interactions on selected material surfaces, so that these parameters may be separated out. Collaborators will supply the range of initial materials. Briefly, processes involving protein adsorption, cell attachment, clustering of cell membrane receptors and assembly of the cytoskeletal complexes will be mapped. Surface characterisation by AFM, SEM, ESCA, TEM, XRD, ETIR will be performed on these surfaces. Identified parameters will be pursued by surface modifications performed in house or by the Industrial Collaborators. These results will provide vital data for the prosthesis manufacturers. A rapid screening technique will be developed to provide manufacturers with the essential feedback they require for optimising and modifying surfaces.
