Manzamine A is a member of a family of alkaloids of marine origin, which have been shown to possess significant anti-tumour and anti-bacterial activity. The formidable synthetic challenges presented by the structural complexity of the manzamines, coupled with their significant biological activity, has stimulated many synthetic endeavours towards the preparation of sub-units of these alkaloids.The aim of the work outlined in our proposal is the efficient synthesis of manzamine A by a strategy which is conceptually novel and employs new metal-catalysed reactions for ring construction. The pivotal transformations in our route will be the tandem catalytic generation and rearrangement of an ammonium ylide to establish the ACE ring system, and a carbenoid C-H-insertion reaction to construct the D ring and complete the tetracyclic core framework.
