A preclinical investigation has revealed that the mycalamides isolated from marine sources are amongst the most potent agents active against human solid tumours discovered to date. They are also active against viruses (Herpes and Poliomyelitis). A synthesis of the mycalamides launched in 1991 will be completed and extended to the theopederins, only discovered in 1992, which show similar activity but benefit from diminished cytotoxicity. Owing to the difficulty of isolating the natural compounds from marine sources, organic synthesis will be essential to elucidating the mode of action and toxicology of these promising new agents.
