This request funds to purchase a Cryostream Nitrogen Gas Cooling Device, to be used in conjunction with a 4-circle X-ray diffraction, for the collection of low temperature single crystal diffraction data. A number of exciting and challenging projects are underway, which all require low temperature structural characterisation facilities. Crystallographic support is required for molecular recognition studies on choramphenicol acetyltransferase and DNA damage, synthetic programmes for chiral taxoids from carbohydrates, organic and organometallic chemistry in asymmetric catalysis, nitrogen-bridged organic systems, and aza- and oxotrimethylenemethane complexes. Precise molecular geometry parameters are needed for studies on metal complexes with unsaturated organic molecules, metal co-ordination compounds, chiral metal complexes and hydrogen-bonding interactions in such systems, and for molecular modelling of reagents to achieve reagent-controlled asymmetric induction. Low temperature studies are required to counter problems of high reactivity and high thermal motion/disorder in inorganic metal fluorides and metal complexes with fluorine-containing ligands. A new programme will be initiated to measure electron density distributions on key compounds connected with the above research programmes.
