In the first year of this project the post-doc, 50% technician and a research student will study and optimise the closed system with vacuum extraction, as described. Initially studies will concentrate on characterising and optimising (eg. minimising retention time and the time required for anaerobic adaptation and maximising carbon substrate utilisation) the aerobic regeneration stage using synthetic media based on the known composition of the anaerobic stage. Subsequently, a complete closed system with vacuum extraction will be set up and characterised, providing data for modelling studies. Key considerations will be optimising volumetric ethanol productivity, maximising active cell retention time in the anerobic stage and minimising the selection pressure for revertant strains. In the second year I.C. will subcontract development work for CAMR (51,000 + vat) where the process will be studied on a larger scale and with a range of feedstocks supplied by AGROL Ltd.
