The objectives of the research programme will be to evolve an efficient and, where possible, inexpensive means of constructing dendritic polysaccharides, ie macromolecules incorporating highly-branched oligosaccharides attached to a central core. In order to achieve this goal, the two challenges that need to be met are (1) the construction of dendron oligosaccharides where each monosaccharide carries two or more branching points, as a result of (a) a convergent step-by-step glycosidation strategy, or (b) the divergent growth of the oligosaccharide chains by the orthoester glycosylation-polycondensation technique, and (2) the development of an efficient procedure for the attachment of branched oligosaccharide dendrons to suitable cores. This objective is probably most simply achieved by the construction of dendrimers containing no more than first generation dendrons. Thus, the attachment of branched dendrons possessing terminal mono- or oligo-saccharides with an appropriate functional group to various central cores will be studied. As soon as the dendritic polysaccharides have been synthesised, they will be characterised and evaluated in the fullest possible manner.
