Linear arrays of 256 separate microelectrodes will be used to electrochemically generate a 256 x 256 array of octamer nucleotides in an area of 10 cm2. The building up of the different sequences will be computer controlled and fully automated. Two optical methods will be investigated to detect the hybridization of a DNA strand with the library of octamer nucleotides. One will be the conventional fluorescence technique which requires labelling the DNA with a chromophore and then using a photon counting camera. The other technique employs imaging ellipsometry to detect the hybridization of unlabelled DNA by the change in optical average thickness at the individual sites. Both techniques will employ digital image analysis which will facilitate automated detection of hybridization, and real time studies of the hybridization process itself. This work will lead to miniature and rapid sequencing procedures for DNA which should greatly assist the human genome project.
