The programme of research seeks to understand the key physicochemical factors that determine the fate of nanosized colloidal carriers within the vascular and lymphatic circulatory systems in the human body. Attention will be focused on particle size in sub 150 nm scale and surface structures at 1-10 nm level (eg attached polymeric groupings). Biodegradable particles with defined surface structures will be prepared by the adsorption and chemical attachment (grafting) of defined structures comprising custom made polymers containing polyethylene oxide moieties. The ability of such particles to control the uptake of plasma proteins will be determined. The critical surface properties of the particles will be characterised by established and novel methods. Nanoparticles with appropriately engineered surfaces will be used to target particles to defined body sites accessible via the vascular and lymphatic circulatory systems; namely the general circulation itself, bone marrow, tumours (and sites of inflamation), spleen, liver and lymph nodes. Biological studies will include studies with cell cultures and animal models.
