In the proposed study the nature of the biochemical and physical environment required for endothelial cells to produce a non-thrombogenic monolayer on a vascular model would be defined, and the nature of blood cell activation and attachment during exposure to flowing blood. The beauty of this approach is that it negates the need to predict the complex interactions between endothelial cells and smooth muscle cells, and the seemingly controlling nature of extracellular matrix. Instead, response of human endothelial cells to well-defined haemodynamic stresses representative of the cardiovascular system, both steady and pulsatile, and under different wall shear rates will be assessed both biochemically and histologically. The nature of the interaction of the cells with blood at a cellular and molecular level will be studied so as to identify the nature of the local physical, biochemical and haemodynamic environment which supports natural endothelial growth.
