The advances in optical microscopy and related optical systems mean that the interpretation of the images formed even by quite simple objects has become increasingly difficult. For the more complex objects, such as those used in the materials and engineering sciences (bulk liquid crystal displays or synthetic or natural polymers) or in the biological sciences (microtubules), the interpretation and understanding of images has become almost impossible without the aid of theoretical description of the light interaction with the specimen. The aim of the present proposal is to provide the necessary theory capable of dealing with anisotropic and inhomogeneous objects of interest by giving a full vectorial treatment of light/specimen interation in optical microscopes and conoscopes. My recent results serve as the basis of proposed programme.This research proposal sets out a programme of a systematic research project aiming at the structures. Specifically the interaction of light with the following types of specimens will be studied; light scattering by large spherical objects, partial coherence effects in fluorescence confocal and conventional microscopes and light interaction with anisotropic specimens. It is anticipated that even for the most complex case of light/specimen interaction the theory will remain relatively uncomplicated, which will make it attractive to scientists from a wide range of disciplines.The research program is designed to closely collaborate with the experiment so that the theory is made in conjunction with experimental work such that theoretical developments can be continuously refined and their significance demonstrated. As a result of the proposed research the currently available microscope and conoscope imaging theories are to be advanced such that the presently not or difficulty interpretable images will provide additional information on the specimen. It is also anticipated that the acquired knowledge will help envisaging newer and more efficient experimental methods to extract additional information concerning the specimen.
