Strains of multidrug resistant (MDR) bacteria are arising which are exceptionally difficult to treat and erradicate from the clinical environment. Foremost of these is methicillin-resistant Staphylococcus aureus which produces MDR proteins that export an array of clinically important antibiotics, including the tetracyclines, fluoroquinolones, macrolides and from the important point of containment, quaternary ammonium antiseptics. The need for new antibiotics and resistance modifiers was compounded in 2002 with the first report of a fully vancomycin-resistant MRSA, this agent being the treatment of choice for MRSA infections. We have evaluated plant natural products that inhibit bacterial resistance mechanisms and one class of these, the julicines are also antibiotic. This dual mode of action has not been described before. We are therefore seeking funding to fully characterise the major active component (julicine A) and minor structurally related metabolites from the producing organism. Additionally, the active compounds from further plant extracts that modify bacterial resistance will be evaluated. Purified natural products will be assessed for efflux inhibition activity against a panel of MRSA strains possessing MDR efflux systems using the requested spectrofluorimter.
