This study will use synthetic ligament scaffolds to investigate the effect of mechanical stimulus (in the form of cyclic strain) on the metabolism of synovial fibroblasts. These scaffolds will be seeded with bovine synovial fibroblasts and incubated in culture medium for predetermined periods, then subjected to cyclic tensile strains similar to what the anterior cruciate ligament experiences during ambulation. Proliferation of cells, their biosynthesis of collagen, proteoglycans and TIMPS will be monitored at 1, 3 and 7 days. Contribution of induced tissue to scaffold's mechanical strength will be quantified at 35 days. The study aims also to establish the strain threshold at which cell activities are triggered. The investigation will shed light on the process of tissue induction around a ligament implant (whether autogenous or synthetic) and will therefore be useful in defining appropriate graft/implant mechanical characteristics and in specifying effective exercise regimes for postoperative rehabilitation of recipients of ligament reconstruction.
