This proposal presents three novel types of methodology which relate to fragmentation reactions to form medium-sized nitrogen,heterocycles. The new reactions should have general applicability to medium-ring amines and amides and are based on (i) modified Schmidt reactions of azides and (ii) fragmentative rearrangements of nitrone triflates and (iii) electron-transfer reactions of azides. The aim is to determine the scope of these new processes and to apply them to the synthesis of a key compound, cleavamine 23, the parent compound forthe Northern fragment of vinblastine and related clinically important anti-cancer drugs.
