There is an extensive literature in the medical journals showing that CO has an important role in the cardiovascular system. Funded by an EPSRC/MRC Discipline Hoppers award, we have shown that [Ru(CO)3CI(glycinate)] releases CO rapidly under physiological conditions. We have demonstrated that [Ru(CO)3CI(glycinate)] suppresses rejection of transplanted hearts, protects isolated hearts from damage as a consequence of ischemia, dramatically increases the rate of recovery of an isolated heart following storage at 4C and suppresses macrophage activity.The proposed research is directed to identifying metal carbonyls which release CO over a period of between 30 minutes and several hours in vivo and can be given to the patient. A range of water soluble carbonyls of manganese, iron and ruthenium containing biologically safe ligands will be synthesised, and their rates of CO loss in the presence of biologically relevant reagents measured at 37C. Suitable compounds will be supplied to collaborating medical researchers for screening for vasodilation, macrophage inhibition and effect on blood platelet aggregation. It is anticipated that such compounds will be active in the cardiovascular svstem.
