Biocatalysts normally become inactivated during use, but the reason is usually unknown. We aim to develop methods to determine the cause of activ loss, as a basis for rational plans to improve stability. The methods will cover: 1) high yield recovery of adsorbed proteins from biocatalysts without (further) covalent modification: 2) identification of covalent changes in recovered enzyme molecules, using the approaches of proteomics (electrophoresis, mass spectroscopy and chromatography), but developing new interpretation strategies appropriate to modifcations found with biocatalysts; 3) detection and characterisation of non-covalent conformational changes in proteins in situ in biocatalyst particles by appropriate physic techniques (fluorescence, infra-red and circular dichroism) and protease susceptibility; 4) recovery of peptides for analysis from covalently attached proteins, using defined and limited cleavage. This is a flagship project of the Pro-Bio Faraday Partnership.
