A major drawback of [1,3]-Bipolar cycloadditions is their inability to provide single products when applied to dipolarophiles which are not significantly biased either electronically or sterically. The present proposals are aimed at addressing this shortcoming by preparing intermediates related or identical to those available from such dipolar cycloadditions but employing 5-endo cyclizations of allylic or propargylic hydroxylamines or hydrazines. By their very nature, such cyd'izations will be regiospecifia Further, these would allow the generation of optically pure products, as the design of substrates will allow the incorporation of optically pure stereogenic centres. Part of this approach work also features the development of new reagents capable of effecting electrophilic amination; this chemistry should find applications in many areas beyond the present studies. In order to validate the model studies, the new chemistry will be applied to a range of targets, chosen in order to demonstrate its positive features, especially with respect to existing approaches which employ such [1,3]-Bipolar cycloadditions as key steps.
