The proposed project is based on the hypothesis that by quantifying the forces which have to be applied to progressively stiffer matrices to elicit a specific cellular response we can calibrate resident cells as cytosensory guages to detect criticalstrain exposure . The aim of the project is to develop a formula whereby molecular outputs from resident cells within a tissue or bioartificial matrix of known material properties can be used to monitor the critical strain accross cells. This will be achieved by applying similar mechanical loads to cells in progressively stiffer matrices and calibrating molecular and protein outputs for matrix deposition/remodelling.
