Modification of natural tissue matrices to implant as acellular constructs, or to recellularise in vitro and re-implant as living tissues is an attractive approach for soft tissue replacements such as ligaments, tendons, skin and heart valves. Penetration of viable cells throughout the matrix during recellularisation in vitro or in vivo remains a challenge.We propose to investigate the use of ultrsonic energy to modify the structure of soft tissue matrices to enhance cellular penetration and recellularisation . A range of ultrasonic energy levels and doses will be studied and the resulting changes in tissue structure , cellular penetration and rate of recellularisation investigated under passive and biomechanically active conditions in vitro.
