The research centres on the application of the permanganate oxidation of 1,5,9-trienes, developed in our group, to the synthesis of complex, biologically active natural product targets cis-panatellin and spinencin. An asymmetric version of the oxidation will be developed and used to prepare a key fragment which will form the core of both targets. The use of chemo- and stereoselective methodology will be emphasised in order to minimise the need for extensive protecting group and functional group manipulations, thus streamlining the synthesis.The work will provide the basis for a general approach to many of the biologically active Annonaceous acetogenins, consequently answering stereochemical ambiguities and ultimately providing access to scarce material for biological evaluation. (We have identified individuals willing to carry out the cytotoxicity studies on our synthetic materials).We have also developed a new asymmetric phase-transfer catalysed oxidative cyclisation reaction, and if time permits, we will investigate its application in the synthesis of Annonaceous acegenins.
