Traditional histological methods for mapping the distribution of trace elements in organs are lacking selectivity, sensitivity and spatial resolution. Laser ablation technology coupled to metal detectors such as ICP-MS have been shown to achieve trace element mapping sub-ppm level with the resolution of better that 100 um when for example, minerals and glasses have been analysed. So far no attempt has been made to try this technology for the mappiong of trace elments in soft tissues. In order to prevent alteration of the microstructure, the tissue must be solidified and sufficiently cooled to achieve reproducible ablation. This proposal is about building a prototype of a cryogenic cell for the ablation process and testing the performance with different types of soft tissue from arsenic-containing organs, which have been analysed by traditional methods.
