This proposal concerns a class of lipids, the phosphoinositides, which although comprising only a small percentage of membrane lipids, play a central role in a number of cellular processes. In particular these lipids play a central role in two key signal transduction pathways. It is generally assumed that the phosphoinositides are not homogeneously distributed in the membrane but rather that they form lipid patches through lateral phase separation. This multidisciplinary and collaborative project will focus on the phase behaviour of these highly polar, highly charged compounds and their miscibility with other key membrane lipids. Two complementary approaches will be employed, first an examination of monolayer behaviour and second studies on bulk lipid mesophases and their dispersions (liposomes). The project aims to (i) determine whether phopshoinositides are localized in domains within membranes; (ii) determine the effects of miscibility on membrane structure; and (iii) examine charge, counterion and pH effects on the phase behaviour of these lipids. The results will have implications both in structural surface chemistry, biophysics and, more generally, surfactant behaviour.
