It is proposed that chirallithium amide bases may prove useful in desymmetrisation reactions of certain types of cyclic diketones. The chemistry will embrace a range of types of diketone, including various ring sizes and substitution patterns, and should provide very useful access to many types of chiral product. We aim to include as substrates for this chemistry 1,3-diketones having at least one hydrogen at the 2-position. For such systems the chiral base chemistry will involve the intermediacy of chiral dianions, which have been scarcely examined.A major part of the project will involve examining the chemistry of non-racemic products that can be obtained from this novel approach, particularly with respect to discrimination between similar ketone functions in the initially-formed products.Chiral products formed in high enantiomeric excess will be of special interest if they constitute potential intermediates in routes to target molecules. In this respect a potential total synthesis of the Madindoline natural products is proposed, which plans a concise and interesting approaches to key diketones, substituted with an appropriate indole, along with methods for their elaboration to the natural products.
