The potential of integrated optical surface plasmon resonance (SPR) sensors to combine electrochemical measurements with sensitive real-time label-free optical detection render them ideal for the study of immobilised films on solid supports and realisation of multisensor arrays with very small sample volume for applications in medical screening, drug discovery and the life sciences. The purpose of this project is to realise novel waveguide SPR-based sensor arrays in microflow systems and demonstrate their combination with electrochemically-controlled localisation of lipid sensing layers on array elements, followed by combined electrochemical and optical measurements of the activity of phospholipase enzymes in the catalysis of lipid membrame reactions, as an example application. The sensor arrays will be applied to clinical samples to study their operation in real matrices. The phospholipase A2 (sPLA2) group of enzymes play a central role in the inflammatory response to infection and trauma, and these studies of enzymes acting at a phospholipid interface will contribute to the understanding and treatment of disease. Novel multifunctional sensor devices will be provided for a range of life science applications.
