Solution structure and dynamics of polypeptides is a topic of intense current interest. However, there is still a dearth of information on several key aspects. We propose to apply the novel spectroscopic technique of Raman optical activity (ROA), which measures vibrational optical activity by means of a small difference in the intensity of Raman scattering from chiral molecules in right and left-circularly polarised incident light, to the problem. On account of its sensitivity to chirality, ROA is a more incisive probe of peptide backbone conformation than conventional Raman and infrared spectroscopy and reveals more details than UVCD. These include loop and turn structure as well as secondary structure in native proteins, together with different states of hydration, even in the coat proteins of intact viruses. A range of polypeptides in model conformations will be studied in both aqueous and non-aqueous solvents. Temperature-dependent ROA measurements will provide information about dynamic aspects of structure. The aim is to obtain new information about important details of solution structure and dynamics that has eluded other techniques and to use the results in the application of ROA to ongoing studies of solution structure, function and misfunction of proteins and to structural virology.
