The research will be directed to the development of a microscale, addressable system for the study of carbohydrate-protein interactions. Carbenes generated by photolysis of a trifluromethyl-substituted diazirine will be used to generate (i) a dextran-coated surface using a diazirine-decorated dendrimer and (ii) a microarray of patches consisting of specific carbohydrates attached to the dextran surface via a photonded dendrimer. The attachment of specific lectins (verotoxin 1 and daffodil lectin) to the engineered surfaces will be studied using fluorescence-tagging proteins. The diazirine-decorated dendrimer will represent a universal polymerisation agent capable of polymerising any organic substance or mixture of substances, and of bonding virtually any organic substance of either low or high molecular weight to an organic polymer surface. The basic phototattachment system will have the potential for extension to the study of many other biological interactions.
