Previous studies by the principal applicant have demonstrated the feasibility of constructing complex, functional molecular architectures based upon self-assembled monolayers (SAMs). The effectiveness of SAM photolithography has also been clearly demonstrated, and photopatterned SAMs have been employed to create micropatterned molecular and cellular materials. However, further progress is prevented by the inherent limitation on feature size imposed by conventional photolithography. The present proposal seeks to open the way to fabrication of a new generation of molecular nanostructures, by exploiting the remarkable resolution of the scanning near field optical microscope (SNOM). Using SNOM photolithography, nanoscale molecular features will be formed and functionalised, and characterised using a range of powerful techniques. They will be employed as resists and the possibility of using them in nanoscale diagnostic tests (in nanoscale immunoassays and DNA analysis) will be demonstrated.
