Summary on Grant Application Form |
A new structurally related series of cationic lanthanide complexes will be prepared whose absolute configuration is controlled by remote stereogenic centres and whose local helicity may be tuned by variation of the Ln ion. The nature of the interaction of these complexes with dodecamer oligonucleotides will be defined by X-ray crystallography, solution NMT (varying Ln ion to select different relaxation/shifting ability) and CD and CPL Binding affinities will be measured by monitoring luminescence changes as a function of added oligo, analysing data by McGhee/van Hippel methods will site and stereoselectivity achieved through variation of Ln (helicity control), and the structural features of the intercalating moiety. Reactivity studies with Tb complexes will seek selective photooxidation at G sites, the possible hydrolytic action of coordinatively unsaturated complexes will also be examined.
