Our group consists of over 40 PhD and postdoctoral workers, many of whom are supported on Research Council Grants. At present we are using a 16 year of Waters Preparative HPLC system for all our difficult chromatographic separations. This instrument is in constant need of repair and technical support. Some spare parts are no longer available and we are in great danger of a complete system failure. This would severely disrupt our work as no other instruments are available to us for the needs of our large research group. We are therefore requesting funds to purchase a preparative HPLC system to support the many and varied projects underway within our group. We have recently invested in a modern, automated analytical HPLC system for the group without recourse to Research Council funding. To complement this equipment we are seeking funding for a preparative system to carry out separations of intermediates within our synthetic programmes. The system would be configured with automated injection and fraction collection. The equipment would be capable of separations ranging in scale from 100mg to many grams, by repetitive, automated, injections if need be. The requested equipment would underpin our work in many different areas, a selection of these include: - Total Synthesis of Biologically Active Natural Products.- Synthesis of Complex Carbohydrates- Protein-Carbohydrate Recognition in Gene Therapy- Contraceptive/Immunosuppressive Carbohydrates- New Polymer Supported Reagents for Orchestrated Multi-step Synthesis- Asymmetric Induction using p-Allyltricarbonyliron Lactone Complexes- A New C-Glycosidation Procedure via Anomeric Heteroatom to Carbon Rearrangements of Tetrahydropyranyl Ethers
