It is proposed to rebuild and improve the temporal and spatical resolution of a fluorescence lifetime imaging microscope (FLIM) based upon single-photon counting detection. The new instrument will permit two-photon excitation of fluorescence, enhancing greatly the confocal and spatial performance. The use of short pulse excitation will further enhance time resolution, permitting study of the details of photochemical kinetics of sensitisers and molecular probes inside living cells.
