The oligodeoxynucleotide 5'-TACTTACATAAA acts as an antisense molecule inhibiting production of TNF-x. Clinical studies are currently proceeding to investigate the topical use of this material for the treatment of psoriasis.The Liverpool team (Cosstick. O'Neil and Roberts) have experience in the synthesis of nucleotide analogue which exhibit stability towards phosphorylases and nucleases. In this programme we wish to incorporate synthetic nucleotides into the aforementioned dodecanucleotide and test their effectiveness as TNF-x antisense agents. Dr Mark Gumbleton (Cardiff University) has agreed to perform the key biological tests (see later).
