Our programme of research will investigate intramolecular additions of carbon centred radicals to pyridines, quinolines and related heteroaromatic compounds. In particular, we intend to delineate the scope of the reaction with respect to (I) additions to C-2, C-3 and C-4; (ii) five, six and higher ring closures and (iii) the effect of pH and substituents on the mode of cyclisation and efficiency of reaction.Parallel studies will seek to apply the chemistry developed to syntheses of some biologically interesting natural products. Targets include the yohimbine alkoloids (which have been shown to be effective in the treatment of penile erectile dysfunction), camptothecin (which displays cytotoxicity towards human ovarian, colon, lung, gastric and CNS cancers) and schumanniophytine (active against the HIV-IIIB virus and type 1 herpes somplex virus.
