The methods of neutron diffraction and isotopic substitution (NDIS) will be applied to complex solutions containing electrolytes, polyelectrolytes and polypeptides. Experimental data will be used to obtain quantitative structural information on i) the hydration of electrolytes in solution with biologically significant polyelectrolytes such as DNA; this will provide insights into ionic specificity.ii) Water structure in the presence of different ions, and as a function of concentration, and temperature; this will help rationalise the mechanisms which underpin the Hofmeister series.iii) Water and ion hydration structure in the presence of polyelectrolytes and polypeptides, and as a function of temperature; this will be used to evaluate the extent to which water structural perturbations and hydrophobic effects influence polyelectrolyte phase behaviour.Isotopically enriched polypeptides and polyelectrolytes will be synthesised, and used in NDIS experiments to probe directly the aqueous environment of specific sites of the polyion.Neutron quasielectric experiments will be carried out to determine the proton dynamics in aqueous.
