MALDI (Matrix Assisted Laser Desorption Ionisation) mass spectrometry provides a rapid and accurate means of obtaining mass data for macromolecules. It is proposed to investigate from both an experimental and theoretical standpoint the limiting spatial resolution attainable in UV laser MALDI to ascertain its potential for use in a microscopic imaging mode.Work will initially make use of spatially homogeneous matrices prepared by sublimation and also explore new methods for uniform incorporation of analytes to allow spot-size dependance to be investigated for protein analytes.Modelling based on an excited-state thermal isolation scheme will be used to assess theoretical factors contributing to resolution. A preliminary investigation will be made of the direct desorption of protein from tissue.If successful this work could bring benefits in macromolecule analysis for biomedical, bioelectronic and chemical applications.
